چکیده

An attempt was made to substantiate a modori (thermal gel degradation) inducing effect of a latent serine proteinase. The proteinase was purified homogeneously from the sarcoplasmic fraction of threadfin bream muscle using a combination of DEAF-cellulose, Con A-Sepharose, ArgSepharose, Shim-pack HAC, and TSK-gel G3000SWXL column chromatographies. Its molecular weight was estimated as 77,000 by SDS-PAGE analysis and 70,000 by gel filtration using TSK-gel G3000SWXL. To examine the occurrence of modori-phenomenon, sols prepared from threadfin bream myofibrils lacking in modori-causing property were heated at 60•Ž in the absence or presence of the proteinase. Only in the presence of the proteinase, marked modori was observed by the puncture test and sensory tests for estimating the physical property of resulting gels, and breakdown of myosin heavy chain was also observed by SDS-PAGE analysis. This evidence clearly suggests that the proteinase is responsible for the modori-phenomenon occurring at 60•Ž.

Latent S49P neuroserpin forms polymers in the dementia familial encephalopathy with neuroserpin inclusion bodies.

The serpinopathies result from conformational transitions in members of the serine proteinase inhibitor superfamily with aberrant tissue deposition or loss of function. They are typified by mutants of neuroserpin that are retained within the endoplasmic reticulum of neurons as ordered polymers in association with dementia. We show here that the S49P mutant of neuroserpin that causes the dementi...

Rapid Synthesis of Nanostructured Pure Anatase TiO2 with High Thermal Stability by Polymeric Sol-Gel Route

The nanostructured anatase TiO2 with high thermal stability was prepared by polymeric sol-gel method without any additives. The particle size distribution of polymeric titania sol was determined by dynamic light scattering (DLS). Then properties of obtained titania were investigated by TG-DTA, XRD, FESEM and TEM. Also, the decolorization capability of resultant anatase was evaluated using methy...

Degradation of myofibrillar proteins by trypsin-like serine proteinases.

The effects of the Ca2+-activated cysteine proteinase, the rat trypsin-like serine proteinase and bovine trypsin on myofibrillar proteins from rabbit skeletal muscle are compared. 2. Myofibrils that had been treated at neutral pH with the Ca2+-dependent proteinase and with the rat enzyme were (a) analyzed by sodium dodecyl sulphate/polyacrylamide-gel electrophoresis and (b) examined in the elec...

The inactivation of native enzymes by a neutral proteinase from rat intestinal muscle.

1. The solubilization and partial purification of a proteinase from the intestinal smooth muscle of rats fed on protein-free diets are described. 2. It has a mol.wt. of about 33000 and it is stable over a narrow pH range. 3. From its susceptibility to known modifers of proteolytic enzymes, it appears to be a serine proteinase of a trypsin-like nature. Active-site titration with soya-bean trypsi...

EXPRESSION OF HUMAN PROTEINASE 3 IN CHINESE HAMSTER OVARY CELLS (CHO-CELLS)

Proteinase 3(PR3) is a human polymorphonuclear leukocyte serine proteinase and is the main target antigen for antineutrophil cytoplasmic antibodies (ANCA) found in Wegener's granulomatosis (WG). We developed a stable expression system for conformationally intact recombinant PR3 (rPR3) in Chinese hamster ovary cells (CHO-cells). The part of PR3 cDNA that encoded the active form of PR3 was s...
